Agilent Technologies, Inc. has detailed a new method for the comprehensive structural analysis of the anticoagulant nadroparin. This approach utilizes the Agilent 1290 Infinity II Bio 2D-LC system coupled with the Agilent 6546 LC/Q-TOF mass spectrometer.

The method is designed to characterize low molecular weight heparins (LMWHs), which are complex biomolecules commonly used as anticoagulant drugs. The two-dimensional liquid chromatography separates different degrees of polymerization (DPs) by size exclusion chromatography in the first dimension, followed by ion-pairing chromatography to resolve different oligosaccharide compositions with the same DP in the second dimension.

High-resolution mass spectrometry provides the capability to resolve isotopic patterns, enabling the identification of up to 28 distinct oligosaccharide compositions within nadroparin. This level of detail highlights the structural diversity of LMWHs and underscores the need for sophisticated analytical techniques.

The developed analytical workflow aims to ensure drug quality and safety. It addresses the structural variations inherent in LMWHs due to different manufacturing processes. This precise characterization is crucial for quality control in the pharmaceutical industry, including the verification of equivalence for generic nadroparin products.
