Researchers have employed the AI protein-folding software AlphaFold to improve the safety of gene-editing proteins. In a recent study published in Nature, the AI was modified to identify key areas within these proteins responsible for off-target DNA edits.

One significant challenge in the development of gene-editing systems has been safety. While these systems can be made highly specific to target desired gene sequences, the vastness of the human genome means that even rare DNA sequences can appear multiple times by chance. This can lead to unintended edits at incorrect locations in the DNA.

Although the probability of an off-target edit occurring in a single instance is low, therapeutic applications require editing a large number of cells, making errors potentially inevitable. The study focused on modifying the identified problematic protein regions to significantly reduce these off-target effects.

The development of safer gene-editing proteins could accelerate the broader adoption of gene therapies by providing greater control over the risks associated with unintended genetic modifications.
