The Alzheimer's Association has initiated the PROTECT-Cog Study, a significant global clinical trial aimed at evaluating whether a combination of a proven multidomain lifestyle intervention and a metabolism-targeting drug, such as a GLP-1 agonist, can reduce the risk of cognitive decline, mild cognitive impairment (MCI), and dementia in at-risk older adults.

The $100 million initiative was announced at the Alzheimer's Association International Conference (AAIC) 2026 in London. This study builds upon the success of the U.S. POINTER study and represents the next phase in research focused on risk reduction and prevention strategies for dementia.

"PROTECT-Cog builds directly on what we learned from U.S. POINTER and takes the next critical step in prevention science," said Maria C. Carrillo, Ph.D., Alzheimer's Association chief science officer. "By testing a combined approach that targets both lifestyle and biology, we have the opportunity to better understand how to meaningfully reduce the risk of cognitive decline before symptoms begin."

The three-year study will enroll older adults at increased risk for cognitive decline, comparing lifestyle program approaches and assessing the impact of adding a metabolism-supporting drug. The goal is to identify the most effective intervention combination for delaying mild cognitive impairment while also evaluating effects on frailty, quality of life, and overall health.
