AstraZeneca: Lynparza and Imfinzi improve response duration in endometrial cancer
New data from AstraZeneca's DUO-E trial shows Lynparza and Imfinzi combination therapy more than doubled median duration of response versus standard chemotherapy in mismatch repair proficient advanced or recurrent endometrial cancer.

AstraZeneca announced updated results from the DUO-E Phase III trial, indicating that its Lynparza (olaparib) and Imfinzi (durvalumab) combination therapy demonstrated significant clinical benefit in patients with advanced or recurrent endometrial cancer.
The analysis focused on patients with mismatch repair proficient (pMMR) tumors. In this subgroup, the median duration of response (DoR) for patients receiving Lynparza and Imfinzi was more than double compared to those receiving chemotherapy alone (18.7 months vs. 7.6 months).
The Lynparza and Imfinzi regimen also showed a 32% reduction in the risk of second progression or death (PFS2) compared to the control arm. Imfinzi monotherapy demonstrated consistent benefits across all secondary endpoints, with the greatest impact observed in patients with mismatch repair deficient (dMMR) disease.
These findings, presented at the 2024 Society of Gynecologic Oncology (SGO) Annual Meeting, highlight the potential of the Lynparza and Imfinzi combination, particularly for the approximately 80% of patients with pMMR advanced or recurrent endometrial cancer who have faced limited treatment advancements.
The results reinforce the potential of both Imfinzi and the Lynparza and Imfinzi regimen as important treatment options for patients with endometrial cancer.
