AstraZeneca's Gefurulimab Shows Improvement in Generalised Myasthenia Gravis Trial

AstraZeneca announced positive results from its PREVAIL Phase III trial, indicating that its drug gefurulimab significantly improved symptoms in adults with generalised myasthenia gravis (gMG). The study met its primary endpoint, showing a statistically significant and clinically meaningful improvement in the Myasthenia Gravis Activities of Daily Living (MG-ADL) score at week 26 compared to placebo in patients with anti-acetylcholine receptor (AChR) antibody-positive gMG.

The trial also met all secondary endpoints, including improvements in the Quantitative Myasthenia Gravis (QMG) total score at weeks four and 26. Notably, clinically meaningful improvements in daily activities were observed as early as week one and were sustained through week 26, suggesting rapid and lasting efficacy. Improvements in muscle strength, measured by the QMG score, were also seen early and maintained throughout the study period.

Gefurulimab is a novel, dual-binding nanobody designed for self-administration via subcutaneous injection. It represents a potential new treatment option for patients with AChR-Ab+ gMG, a rare autoimmune disorder characterised by muscle weakness and loss of function. The company stated that the drug was generally well-tolerated, with a safety profile consistent with previous C5 inhibitor trials, and most adverse events were mild to moderate.

Myasthenia gravis can cause debilitating and unpredictable symptoms, affecting patients' quality of life. The results from the PREVAIL trial reinforce the potential of gefurulimab to offer a convenient and effective treatment option, addressing the significant unmet needs of patients with this condition.
