AstraZeneca's Truqap combination improves progression-free survival in advanced prostate cancer
AstraZeneca announced that its drug Truqap, in combination with abiraterone and hormone therapy, demonstrated a statistically significant improvement in radiographic progression-free survival for patients with PTEN-deficient metastatic hormone-sensitive prostate cancer.

AstraZeneca's CAPItello-281 Phase III trial has shown that Truqap (capivasertib) in combination with abiraterone and androgen deprivation therapy (ADT) significantly improved radiographic progression-free survival (rPFS) for patients with PTEN-deficient metastatic hormone-sensitive prostate cancer (mHSPC). The combination was compared against abiraterone and ADT with placebo.
This marks the first time an AKT inhibitor combination has demonstrated a benefit in this specific subtype of prostate cancer. PTEN-deficient mHSPC is an aggressive form of the disease with historically poor outcomes. Approximately one-quarter of the estimated 200,000 annual mHSPC diagnoses fall into this patient group.
Overall survival (OS) data were immature at the time of analysis, but the Truqap combination showed an early trend towards OS improvement. The trial will continue as planned to further assess OS as a key secondary endpoint.
AstraZeneca intends to present the full data at an upcoming medical meeting and submit it to global regulatory authorities. The safety profile of Truqap in combination with abiraterone and ADT in CAPItello-281 was consistent with the known profiles of each medicine.
