Biomarker Technologies (BMKGene) has detailed a methodology to improve the reliability of variant detection in whole exome sequencing (WES) through the application of unique molecular identifiers (UMIs). The process is designed to mitigate common technical errors introduced during library preparation and PCR amplification.

In standard exome sequencing, distinguishing between low-frequency somatic variants and background sequencing noise is a recurring challenge. By incorporating UMIs, researchers can track individual DNA molecules, allowing for more precise deduplication and error correction. Field Application Specialist Catalina Alvarez noted that this molecular barcoding provides a clearer distinction between biological variants and artifacts caused by the sequencing process.

BMKGene currently provides human whole exome sequencing services utilizing the Agilent SureSelect capture strategy. The company emphasizes that while UMI integration provides significant improvements in data confidence, the final analytical performance remains dependent on high-quality input samples, sufficient sequencing depth, and robust bioinformatics pipelines.
