Biomarker Technologies provides guidance on HMW DNA handling for genomic sequencing
BMKGene has released technical guidance on handling high molecular weight DNA to improve PacBio HiFi genome assembly outcomes.

Biomarker Technologies (BMKGene) has published technical guidance regarding the preparation of high molecular weight (HMW) DNA for PacBio HiFi genome assembly. The integrity of DNA fragments remains a critical factor in achieving high-quality genome assemblies in long-read sequencing workflows.
Ana Vila-Santa, Field Application Specialist Manager at BMKGene, noted that long DNA molecules are highly susceptible to mechanical damage. The company advises researchers to minimize vigorous handling, such as vortexing and repeated pipetting, to prevent the fragmentation of DNA strands, which are essential for bridging repetitive genomic regions.
Beyond physical handling, the company underscores that DNA concentration alone is insufficient for assessing sample suitability. BMKGene recommends verifying integrity via gel electrophoresis to ensure that samples meet the stringent requirements of long-read sequencing technology.
This guidance is part of the company's efforts to support its de novo genome sequencing services. Researchers are encouraged to consult with BMKGene’s technical team regarding sample preparation protocols prior to project submission.
