The complement C1 component is a crucial part of the classical complement pathway, typically activated by antibody recognition or pathogens. C1 inhibitors target enzymes within this pathway, such as C1r and C1s, and are being investigated for the treatment of various conditions. Therapies under development aim to reach stages that could offer new therapeutic options for complement-mediated diseases.
The report highlights Sutimlimab from Bioverativ, a monoclonal antibody in the preregistration stage targeting the C1s protein. This treatment is intended to block the classical complement pathway's function, potentially reducing erythrocyte lysis in conditions like autoimmune hemolytic anemia. Notably, sutimlimab targets only one of the complement system's three arms.
Another area of focus is Annexon's ANX007, a monoclonal antibody fragment in clinical development. It is being studied for complement-mediated neurodegenerative ophthalmic diseases and aims to inhibit C1q binding, thereby downregulating the classical complement cascade. ANX007 is currently in Phase II clinical trials.
