Pfizer Inc. announced that its investigational Group B Streptococcus (GBS) vaccine candidate, GBS6 (PF-06760805), has received Breakthrough Therapy Designation from the U.S. Food and Drug Administration (FDA). This designation is intended to expedite the development and regulatory review of the vaccine, which aims to prevent invasive GBS disease in newborns and young infants through maternal immunization during pregnancy.

The FDA's decision is based on an interim analysis of a Phase 2 placebo-controlled study evaluating the safety and immunogenicity of GBS6 in healthy pregnant women. The study is ongoing, and Pfizer will publish the outcomes upon its completion. A Breakthrough Therapy Designation is granted when preliminary clinical evidence indicates that a drug or vaccine may demonstrate substantial improvement over available therapy on a clinically significant endpoint.

Group B Streptococcus infections can have severe consequences for newborns, including sepsis, pneumonia, and meningitis. While current preventive measures like prenatal screening and intrapartum antibiotics help in developed countries, they are not fully protective, especially in the first week of life, and face challenges in low- and middle-income countries. If approved, Pfizer's GBS6 vaccine could offer a critical public health solution.

GBS6 is a hexavalent conjugate vaccine designed to protect against the six most prevalent GBS serotypes, responsible for 98% of GBS disease worldwide. Pfizer is pursuing a clinical development strategy to make a successfully developed vaccine available globally as quickly as possible, with ongoing trials in South Africa, the UK, and the US.
