IASO Biotechnology (IASO Bio) has presented initial findings from its first-in-human Phase 1 clinical trial of IASO206, a BCMA-targeted in vivo CAR-T cell therapy. The treatment is engineered to generate functional BCMA-directed CAR-T cells within the patient's body following a single intravenous infusion, circumventing the need for ex vivo cell manufacturing and lymphodepleting chemotherapy.

The results were unveiled at the International Myeloma Society (IMS) Annual Meeting. The study involved 10 patients diagnosed with relapsed or refractory multiple myeloma. This patient group had a high prevalence of high-risk and ultra-high-risk cytogenetic features, accounting for 90% and 70% respectively. The trial reported an objective response rate (ORR) of 90% and a minimal residual disease (MRD) negativity rate of 90%. In the cohort that received the highest dose, both ORR and MRD negativity reached 100%.

The safety profile of the therapy was deemed manageable. Adverse events were primarily mild infusion-related reactions, such as fever and chills. Cytokine release syndrome (CRS) was mostly Grade 1 and lasted an average of 2.5 days. No dose-limiting toxicities, neurotoxicity, or treatment-related deaths were reported.

IASO Bio is advancing the clinical development of the therapy, including further evaluation of the optimal dose. The preliminary results from this first-in-human study suggest that IASO206 may offer significant benefits for patients with advanced multiple myeloma.
