Novartis presents JDQ443 data in KRAS G12C-mutated solid tumors
Novartis presented efficacy and safety data for JDQ443, an investigational KRAS G12C inhibitor, in patients with KRAS G12C-mutated solid tumors, including NSCLC, at ASCO 2023.

Novartis shared promising data on its investigational drug JDQ443 at the American Society of Clinical Oncology (ASCO) 2023 meeting, highlighting its activity in KRAS G12C-mutated solid tumors, particularly non-small cell lung cancer (NSCLC).
The KontRASt-01 study, with a data cutoff of February 1, 2023, indicated an objective response rate (ORR) of 57.1% in NSCLC patients at the recommended 200 mg twice-daily dose. The disease control rate (DCR) was reported at 92.9%.
Regarding safety, JDQ443 demonstrated a favorable profile. Treatment-related adverse events (TRAEs) were predominantly low-grade. No Grade 4-5 TRAEs were observed. Nausea, vomiting, and diarrhea did not exceed Grade 2. Elevations in liver enzymes (ALT and AST) were infrequent and transient.
JDQ443 is a covalent KRAS G12C inhibitor designed to irreversibly bind to and inhibit the mutated protein, aiming to disrupt cancer cell signaling. This is particularly relevant for NSCLC, where KRAS G12C mutations are a common driver. Novartis is exploring JDQ443 in both monotherapy and combination studies.
