Pfizer's ADCETRIS Combination Therapy Shows Improved Overall Survival in DLBCL Trial
Pfizer's Phase 3 ECHELON-3 trial demonstrated that a combination therapy including ADCETRIS significantly improved patients' overall survival compared to placebo.

Pfizer Inc. has announced positive results from its Phase 3 ECHELON-3 trial, evaluating ADCETRIS® (brentuximab vedotin) in combination with lenalidomide and rituximab for the treatment of relapsed/refractory diffuse large B-cell lymphoma (DLBCL). The study showed a statistically significant and clinically meaningful improvement in overall survival (OS) when compared to lenalidomide and rituximab plus placebo.
The trial also observed positive outcomes in key secondary endpoints, including progression-free survival (PFS) and overall response rate (ORR). The results were deemed encouraging as they included heavily pre-treated patients, some of whom had received prior CAR-T therapy. The safety and tolerability profile of the treatment was consistent with previously presented data.
Diffuse large B-cell lymphoma is the most common type of lymphoma and an aggressive blood cancer. Approximately 40% of patients do not respond to first-line treatment or experience relapse after initial therapy, highlighting the need for new treatment options.
Pfizer plans to submit the trial data to the U.S. Food and Drug Administration (FDA) to potentially support a regulatory filing. ADCETRIS currently has seven approved indications in the U.S. and has been used in over 55,000 patients since its initial approval in 2011.
