Roots Analysis Identifies Key Companies in Protein Degradation Therapeutics

Roots Analysis released a new report in July 2024 focusing on the targeted protein degradation (TPD) market and its leading companies.
TPD therapies have significantly advanced protein homeostasis by enabling the destruction of disease-causing proteins. While traditional methods like small molecule inhibitors could only target a fraction of pathological proteins, TPD utilizes the body's cellular machinery to degrade previously undruggable targets, potentially addressing over 80% of disease-causing proteins. This approach employs bifunctional molecules that recruit E3 ubiquitin ligases to target proteins, leading to their ubiquitination and subsequent proteasomal degradation.
The report identifies eight key companies with advanced pipelines in TPD. These include Arvinas, AstraZeneca, Bristol-Myers Squibb, C4 Therapeutics, Loxo Oncology, Olema Oncology, Radius Health, and Roche. Arvinas, a clinical-stage biopharmaceutical company leveraging its PROTAC platform, recently received FDA Fast Track designation for its breast cancer drug candidate, ARV-471 (Vepdegestrant), and has a partnership with Novartis for another candidate, ARV-766.
