Sanofi has received orphan drug designation from Japan's Ministry of Health, Labour and Welfare (MHLW) for its investigational drug rilzabrutinib to treat IgG4-related disease (IgG4-RD). This designation is granted to medicines intended for rare diseases or conditions with unmet medical needs. IgG4-RD is a rare, progressive, immune-mediated chronic condition that can cause significant organ damage.

The designation follows positive data from a Phase 2 study where rilzabrutinib demonstrated a reduction in disease flares and the need for glucocorticoid treatment over 52 weeks. The study's safety profile was consistent with previous trials, with no new safety signals identified. Sanofi is currently evaluating rilzabrutinib in a Phase 3 study for IgG4-RD.

Rilzabrutinib is being studied for multiple rare immune-mediated diseases. The drug candidate has previously received similar designations globally for IgG4-RD. It has also received approvals for immune thrombocytopenia (ITP) in the US, EU, and UAE, with a regulatory review for ITP ongoing in Japan.

Rilzabrutinib is an oral, reversible covalent Bruton's tyrosine kinase (BTK) inhibitor designed to restore immune balance. BTK plays a critical role in various immune-mediated diseases and inflammatory pathways. Sanofi continues to explore rilzabrutinib's potential across a range of rare conditions.
